研究领域:
1. 新型分子影像探针
分子影像探针在肿瘤影像诊断和治疗中的研究价值和临床意义正日益凸显。而探针表面蛋白非特异性吸附在很大程度上影响其特异性和敏感性。张兵波博士紧紧围绕上述问题,利用蛋白仿生策略,旨在消除纳米探针的非特异性吸附,削弱免疫系统的摄取,实现对肿瘤有效诊疗。先后开展了纳米颗粒蛋白仿生修饰、蛋白介导生物模拟仿生合成和相关机制及其扩展等一系列研究。主要学术成果包括:(1)借助超声波作用力充分暴露蛋白亚结构域ⅡA与ⅢA疏水腔,对一系列疏水纳米颗粒进行了简洁高效的蛋白仿生修饰,获得了抗非特异性吸附能力极强的纳米探针,用于体内外生物医学成像与检测;(2)在上述研究基础上,利用蛋白功能氨基酸序列生物模拟仿生合成了系列诊疗探针,用于肿瘤单/多模态增强显影和诊疗一体化研究; 发展了一种绿色、简洁且高效的多功能诊疗基元仿生集成新方法;(3)证实了蛋白质中的氨基酸(C、D、E、W、Y)具有仿生合成功能,为探索新型功能蛋白提供了实验基础和理论依据。
2. 体外分子诊断
本课题组注重纳米材料的生物医学应用,且已在这个方面做了大量的研究工作。在体外诊断方面,利用自行合成的纳米材料,经表面工程化修饰,标记相应的抗体分子,对体外血清,尿液等进行特异性,超灵敏检测。利用荧光量子点标记AFP单克隆抗体,结合免疫层析试纸条技术,对1000 例临床血样进行了AFP蛋白的快速、灵敏检测(Biosensors & bioelectronics,2011,30, 145-150);基于量子点具有一元激发多元发射的独特性质,利用自制的多色量子点经行了高通量检测的验证。基于液相生物芯片技术,采用自制的量子点荧光探针和聚苯乙烯微球,对甲胎蛋白(AFP)进行了检测。

科研业绩:
张兵波博士长期致力于生物医学材料和分子影像研究。 迄今以第一或通讯作者在Advanced Materials, Nano letters, ACS nano, Advanced Science, Biomaterials, Theranostics等国际著名期刊共发表SCI论文近50篇(H因子20; IF>10 4 篇;JCR一区26篇);主编英文专著一部(World Scientific, 2016)。 张兵波博士完成及在研主持项目有:国家自然科学基金(3项),上海市教委科研创新重点项目, 同济大学优秀青年培养计划项目,同济大学英才计划(优秀、骨干教师)项目,中央高校基本科研业务费专项资助基金和多项国家及教育部重点实验室研究基金等。 热烈欢迎对生物材料、生物医学工程、体外诊断试剂及便携式仪器开发、分子影像探针等有强烈兴趣的同仁加入本课题小组,共同发展!
